Protocol Outline

1. Prepare all reagents, samples and standards as instructed in the manual. 2. Add 50 µl of standard or sample to each well. 3. Add 50 µl of  Bioaim mix. 4. Incubate 1 h at RT. 5. Wash and add 100 µl of TMB One-Step Substrate Reagent to each well. 6. Incubate 15 min at RT. 7. Add 50 µl of Stop Solution to each well. 8. Read at 450 nm/570 nm.

Background

VEGF (Vascular endothelial growth factor) is a potent mediator of both angiogenesis and vasculogenesis in the fetus and adult. It is required for regulating the proliferation, migration, and survival of embryonic endothelial cells and during wound healing and the female reproductive cycle in adults. Pathologically, it is involved in tumor development and tumor vascular leakage. VEGF binds to VEGF R1/Flt-1, VEGF R2/Flk-1/KDR, and Neuropilin-1. Multiple forms of VEGF are generated by alternative splicing and proteolysis. In addition to binding to VEGFRs, VEGF binds to receptor complexes consisting of both neuropilins and VEGFRs. This receptor complex has increased VEGF signalling activity in endothelial cells. Neuropilins are pleitrophic receptors and therefore other molecules may interfere with the signalling of the NRP/VEGFR receptor complexes.

Storage

The entire kit may be stored at -20°C for up to 1 year from the date of shipment. Avoid repeated freeze-thaw cycles. The kit may be stored at 4°C for up to 6 months. For extended storage, it is recommended to store at -80°C.

协议大纲

1.按照手册中的说明准备所有试剂，样品和标准品。
2.向每个孔中添加100 µl标准液或样品。
3.在室温或O / N在4°C下孵育2.5小时。
4.洗涤并向每个孔中加入100 µl准备好的生物素抗体。
5.在室温下孵育1小时。
6.洗涤并向每个孔中加入100 µl准备好的链霉亲和素溶液。
7.在室温下孵育45分钟。
8.洗涤并向每个孔中添加100 µl TMB一步底物试剂。
9.在室温孵育30分钟。
10.向每个孔中添加50 µl终止溶液。
11.在450 nm / 570 nm处读取。

背景

脂联素，也称为Acrp30，是脂肪细胞衍生的蛋白质，对代谢和炎症具有广泛的旁分泌和内分泌作用。它促进脂肪细胞分化，脂肪酸分解代谢和胰岛素敏感性，并且与肥胖症，2型糖尿病和动脉粥样硬化形成负相关。脂联素缔合成70 kDa非共价连接的三聚体，该三聚体被二硫键连接成中等分子量的六聚体。
